碲纳米片的制备及其在白血病体外治疗中的应用

Preparation of tellurium nanosheets and their application in the in vitro treatment of leukemia

  • 摘要: 目的:在水相中制备碲纳米片(Te NSs),并研究其在体外环境下对白血病细胞HL-60的杀伤作用。方法:通过液相剥离技术制备Te NSs,并通过X射线衍射仪(XRD)、透射电子显微镜(TEM)和原子力显微镜(AFM)等技术对其进行表征,通过CCK-8实验、倒置显微镜以及Calcein/PI荧光染色等手段考察Te NSs对HL-60细胞增殖能力的影响。结果:成功合成Te NSs,TEM图像显示该纳米片的横向尺寸约为100 nm,AFM结果表明其厚度小于20 nm。细胞实验表明,Te NSs能够诱导HL-60细胞凋亡;0.81 g/L Te NSs与HL-60细胞共孵育24 h后,细胞致死率高达85.26%。结论:Te NSs作为一种新型纳米药物,能够有效杀伤白血病细胞,展现了良好的抗肿瘤效应,在白血病的体外治疗中表现出较大的潜能。

     

    Abstract: Objective: To prepare tellurium nanosheets(Te NSs) in aqueous phase and study their killing effect on leukemia HL-60 cells in vitro. Methods: Te NSs were prepared by liquid phase exfoliation technique and characterized by X-ray diffraction(XRD), transmission electron microscopy(TEM) and atomic force microscopy(AFM). The effect of Te NSs on the proliferation of HL-60 cells was investigated by cell counting kit-8(CCK-8)experiment, inverted microscope, and Calcein/PI fluorescent staining. Results: Te NSs were successfully synthesized. TEM images showed that the lateral dimension of the Te NSs was about 100 nm, and AFM results showed that its thickness was less than 20 nm. Cell experiments showed that Te NSs could induce the apoptosis of HL-60cells, and the lethality rate of cells was as high as 85.26% after 24 h incubation of 0.81 g/L Te NSs with HL-60cells. Conclusion: As a new type of nanomedicine, Te NSs can effectively kill leukemia cells and have good antitumor effect, showing great potential in the in vitro treatment of leukemia.
