Preparation of monoclonal antibody against VASN-EGFFN3 recombinant protein

  • 摘要: 目的:通过大肠杆菌表达系统表达和纯化VASN-EGFFNN3 重组蛋白,免疫小鼠制备抗VASN-EGFFN3单克隆抗体。方法:构建pET30a(+)-VASN-EGFFNN3重组载体,以大肠杆菌BL21(DE3)为载体表达VASN-EGFFNN3重组蛋白。用镍离子亲和层析柱纯化VASN-EGFFNN3 重组蛋白。利用纯化后的VASN-EGFFN3 重组蛋白免疫BALB/c 小鼠后,用杂交瘤技术制备抗VASN-EGFFN3单克隆抗体。结果:成功构建pET30a(+)-VASN-EGFFNN3重组原核表达质粒,诱导的重组VASN-EGFFNN3蛋白主要以包涵体的形式存在。纯化蛋白免疫BALB/c小鼠后获得的血清抗体效价大于1∶2 187 000,免疫效果良好。利用杂交瘤技术获得2株杂交瘤细胞株。结论:成功利用大肠杆菌表达系统制备并纯化VASN-EGFFNN3重组蛋白,通过杂交瘤技术成功制备小鼠抗VASN-EGFFN3单抗。

     

    Abstract: Objective:To express and purify VASN-EGFFNN3 recombinant protein by Escherichia coli(E.coli-)expression system and immunize mice to prepare anti-VASN-EGFFNN3 mAb.Methods:A pET30a(+)-VASNEGFFNN3 recombinant vector was constructed. E.coli BL21(DE3) was used as the vector to express VASNEGFFNN3 recombinant protein.The VASN-EGFFNN3 recombinant protein was purified by nickel ion(Ni)affinity chromatography column.After immunizing BALB/c mice with purified VASN-EGFFN3 recombinant protein, anti-VASN-EgffN3 monoclonal antibody was prepared by hybridoma technique.Results:A pET30a(+)-VASNEGFFNN3 recombinant prokaryotic expression plasmid was successfully constructed.The induced recombinant VASN-EGFFNN3 protein mainly existed in the form of inclusion bodies.The antibody titer of serum obtained from BALB/c mice immunized with the purified protein was tested greater than 1:2187000, indicating a good immune effect.Two hybridoma cell lines were obtained using hybridoma technique.Conclusion:The VASN-EGFFNN3 recombinant protein is successfully prepared and purified by using E.coli expression system and the mouse anti-VASN-EGFFN3 monoclonal antibody is successfully prepared by hybridoma technology.
