环状RNA hsa_circ_0001801在先天性心脏病相关肺动脉高压患儿全血中的表达及其临床意义

Expression and clinical significance of circular RNA hsa_circ_0001801 in whole blood of chil⁃dren with pulmonary arterial hypertension associated with congenital heart disease

  • 摘要: 目的:探讨环状RNA hsa_circ_0001801 在先天性心脏病(CHD)相关肺动脉高压(PAH)患儿中的表达及其临床意义。方法:收集30 例CHD 和30 例PAH-CHD 患儿全血样本,并收集14 例健康儿童全血样本作为对照组,采用RT-qPCR 检测hsa_circ_0001801 在不同分组中的表达量,并与临床资料进行相关性分析,通过Logistic 回归分析确定PAH-CHD 的影响因素。结果:PAH-CHD 组hsa_circ_0001801 相对表达量低于对照组和CHD 组(P< 0.05),而CHD 组与对照组无明显差异。Spearman相关分析显示,hsa_circ_0001801表达与肌酸激酶同工酶呈正相关关系(r=0.393,P=0.002)。多因素Logistic回归分析显示,hsa_circ_0001801为PAH-CHD的保护因素(OR=0.465,95% CI:0.257~0.844,P=0.012)。结论:hsa_circ_0001801在PAH-CHD患儿全血中异常表达,具有作为PAH-CHD生物标志物的潜力。

     

    Abstract: Objective:To explore the expression and clinical significance of circular RNA hsa_circ_0001801 in children with pulmonary arterial hypertension (PAH) associated with congenital heart disease (CHD).Methods:The whole blood samples of 30 children with CHD and 30 children with PAH-CHD were collected, and the whole blood samples of 14 healthy children were collected as the control group.The expression levels of hsa_circ_0001801 in different groups were detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR), correlation analysis was performed with the clinical data, and the influencing factors for PAHCHD were determined by Logistic regression analysis.Results:The relative expression level of hsa_circ_0001801 in PAH-CHD group was lower than that in control group and CHD group(P< 0.05), while there was no significant difference between CHD group and control group.Spearman correlation analysis showed a positive correlation between the expression of hsa_circ_0001801 and creatine kinase-MB (CK-MB) (r=0.393, P=0.002); multivariate Logistic regression analysis showed hsa_circ_0001801 was a protective factor for PAH-CHD (OR=0.465, 95% CI:0.257-0.844, P=0.012).Conclusion:hsa_circ_0001801 is aberrantly expressed in whole blood of children with PAH-CHD and has potential as a biomarker for PAH-CHD.
